چکیده
Abstract Regulatory T cells (Tregs) harness a central role in peripheral immune tolerance. Since its discovery, there have been numerous efforts to dissect the of Tregs context disease condition. Recently, Treg fields are moving from academia industry since many evidences accumulated for potential treatment inflammatory by administering genetically engineered Tregs. However, pre-clinical or clinical trials, this rare cell subset needs be expanded ex-vivo condition achieve sufficient number controlling disease. Our group previously established expansion with 25 mer oligonucleotide maintain stability and function non-biased investigations on perturbation transcriptomic epigenetic changes during culture has not explored. cutting edge multiomics tools which can simultaneously profile transcript signatures single resolution commercialized, we adopt 10x multiome technique investigate our frozen Tregs(eTreg) compared freshly isolated human Treg(fTreg) same donor. Compared fTreg cells, identified new that exists only eTreg named it eeTreg. We found eeTreg resembles phenotype both effector tumor-infiltrated is characterized significantly increased expression chromatin accessibility score FOXP3, STAT5B, ETS1 RUNX1, known essential transcription factors regulatory function. In summary, suggest novel treg might manufactured manipulating

CTLA4Ig inhibits effector T cells through regulatory T cells and TGF-β.
The CD28 costimulatory receptor is a critical regulator of T cell function, making it an attractive therapeutic target for the treatment of immune-mediated diseases. CTLA4Ig, now approved for use in humans, prevents naive T cell activation by binding to B7 proteins and blocking engagement of CD28. However, CTLA4Ig suppresses inflammation even if administered when disease is established, suggest...
متن کاملThe Epigenetic Regulation of Blinatumomab Gene Expression: Tumor Cell-dependent T cell Response against Lymphoma Cells and Cytotoxic Activity
Conventional treatment for cancer such as surgical resection and chemotherapy can cause damage in cases with advanced cancers. Moreover, the identification of tumor-specific targets has great importance in T-cell therapies. For decades, T cell activity has been stimulated to improve anti-tumor activity. Bispecific antibodies have attracted strong interest from pharmaceutical companies, for thei...
متن کاملRegulatory T cells modulate staphylococcal enterotoxin B-induced effector T-cell activation and acceleration of colitis.
Oral administration of bacterial superantigen Staphylococcus aureus enterotoxin B (SEB) activates mucosal T cells but does not cause mucosal inflammation. We examined the effect of oral SEB on the development of mucosal inflammation in mice in the absence of regulatory T (Treg) cells. SCID mice were fed SEB 3 and 7 days after reconstitution with CD4(+) CD45RB(high) or CD4(+) CD45RB(high) plus C...
متن کاملContribution of regulatory T cells and effector T cell deletion in tolerance induction by costimulation blockade.
Blocking of costimulatory signals for T cell activation leads to tolerance in several transplantation models, but the underlying mechanisms are incompletely understood. We analyzed the involvement of regulatory T cells (Treg) and deletion of alloreactive cells in the induction and maintenance of tolerance after costimulation blockade in a mouse model of graft-vs-host reaction. Injection of sple...
